Comma is put after "O" if one may misunderstand the meaning. Look at the following examples:
He didn't drink the milk.= O, sütü içmedi. If I don't put a comma here, "O" is an adjective. Which milk? Answer: O(that)milk. But here we put a comma not to be misunderstood.

Don't mention it. But I usually put a comma after "O" to emphasize.
