Se trata de un acento diacrítico como en español:
donde - dónde, cuando - cuándo

που - πού

"Που" sin acento también se puede traducir como "donde" (sin acento) pero puede traducirse de muchas otras maneras (igual que πού con acento). Aquí te dejo unos pocos ejemplos para que veas como se utiliza. Es, por cierto, una palabra de muchísimo uso en griego, viene a ser como el "que" en español.

Con acento:
Πού είναι η μάνα σου; = ¿Dónde está tu madre?
Πού πας; = ¿A dónde vas?
Πού άφησες τα κλειδιά σου; Δε θυμάμαι πού τα άφησα. = ¿Dónde dejaste tus llaves? - No me acuerdo dónde las dejé.
Tον ρώτησαν πού δουλεύει. = Le preguntaron dónde trabaja.
Προσπάθησε να κοιμηθεί· αλλά πού! = Intentó dormir - ¿pero cómo?
Πού το κατάλαβες; (= πώς έγινε και το κατάλαβες;) = ¿Cómo te diste cuenta?

Sin acento:
Να το σπίτι που μένει ο αδερφός μου. = Esa es la casa donde vive mi hermano.
Είμαι εκεί που θέλω να βρίσκομαι. = Estoy ahí donde quiero estar.
Μην ανακατεύεσαι εκεί που δε σε σπέρνουν. = No te metas donde no te llamen.

Tώρα που θα φύγεις, μη μας ξεχάσεις. = Ahora que te marchas, no nos olvides.
Εδώ που τα λέμε.... = μια και πιάσαμε το θέμα = hablando del tema...
Τι βλάκας που είναι! = ¡Qué idiota que es!
Tην είδε που του μιλούσε (= να του μιλά).
Εκείνος που μιλάει είναι ο αδελφός μου. = Aquél que habla es mi hermano.

Como puedes ver, no se limita a una traducción. Espero que te haces una idea aproximada aunque la verdad es que seguramente no resulta muy fácil al principio.
