"zero" is usually used in maths-operations.
I would suggest the first one, if not understood, the second one.
As you said, we should prefer saying "oh" instead of "zero"

But if the person who asked for the room number doesn't speak English well, he or she would more likely hear "zero" instead of "oh". Also, when talking over the phone, "zero" is commonly used in the UK.

Dear Yessoos, preferences are different everywhere.So why not?
But if somebody can ask his/her room number and go to UK should know the numbers.:)
